DNA Evidence of a Croatian and Sephardic Jewish Settlement on the North Carolina Coast Dating from the Mid to Late 1500s
While the British origins of North American colonization currently are widely accepted, there is new evidence that other countries and non-Christians may have been earlier in establishing permanent settlements on the North Atlantic coast. Using the new research tool of human genomics, this paper provides DNA evidence that Croatians and Sephardic Jews were absorbed into the ancestral population of the Lumbee Native American tribe of North Carolina during the mid- to late-1500s. We further propose that these Sephardic Jews originated, in part, from a subgroup of the Roanoke colonists of 1586. Given this, a new historical narrative of early European colonization in North America during the 1500s is proposed
Space-time evolution and HBT analysis of relativistic heavy ion collisions in a chiral SU(3) x SU(3) model
The space-time dynamics and pion-HBT radii in central heavy ion-collisions at
CERN-SPS and BNL-RHIC are investigated within a hydrodynamic simulation. The
dependence of the dynamics and the HBT-parameters on the EoS is studied with
different parametrisations of a chiral SU(3) sigma-omega model. The
selfconsistent collective expansion includes the effects of effective hadron
masses, generated by the nonstrange and strange scalar condensates. Different
chiral EoS show different types of phase transitions and even a crossover. The
influence of the order of the phase transition and of the difference in the
latent heat on the space-time dynamics and pion-HBT radii is studied. A small
latent heat, i.e. a weak first-order chiral phase transition, or even a smooth
crossover leads to distinctly different HBT predictions than a strong first
order phase transition. A quantitative description of the data, both at SPS
energies as well as at RHIC energies, appears difficult to achieve within the
ideal hydrodynamical approach using the SU(3) chiral EoS. A strong first-order
quasi-adiabatic chiral phase transition seems to be disfavored by the pion-HBT
data from CERN-SPS and BNL-RHIC
Syllable Weakening in Kagoshima Japanese An Element-Based Analysis
This paper examines syllable weakening or nisshōka (入声化) in Kagoshima Japanese (KJ), where high vowel apocope feeds lenition, leading to correspondences such as Tōkyō Japanese (TJ) [kaki] ‘persimmon’ and Kagoshima [kaʔ]. The traditional pattern noted in the literature is quite clear. Apocope elides stem-final /u/ or /i/. The preceding onset is lenited in one of four ways: 1) stops and affricates are debuccalised (/kaki/ > [kaʔ] ‘persimmon’); 2) fricatives undergo voicing neutralisation (TJ [kazu] > KJ [kas] ‘number’); 3) nasals undergo place loss (TJ [kami] > KJ [kaɴ] ‘paper’); 4) rhotics undergo gliding (TJ [maru] > [maj] ‘round’). Dynamics of Hot Bulk QCD Matter: from the Quark-Gluon Plasma to Hadronic Freeze-Out
We introduce a combined macroscopic/microscopic transport approach employing
relativistic hydrodynamics for the early, dense, deconfined stage of the
reaction and a microscopic non-equilibrium model for the later hadronic stage
where the equilibrium assumptions are not valid anymore. Within this approach
we study the dynamics of hot, bulk QCD matter, which is expected to be created
in ultra-relativistic heavy ion collisions at the SPS, the RHIC and the LHC.
Our approach is capable of self-consistently calculating the freeze-out of the
hadronic system, while accounting for the collective flow on the hadronization
hypersurface generated by the QGP expansion. In particular, we perform a
detailed analysis of the reaction dynamics, hadronic freeze-out, and transverse

Diquat Derivatives: Highly Active, Two-Dimensional Nonlinear Optical Chromophores with Potential Redox Switchability
In this article, we present a detailed study of structure−activity relationships in diquaternized 2,2′-bipyridyl (diquat) derivatives. Sixteen new chromophores have been synthesized, with variations in the amino electron donor substituents, π-conjugated bridge, and alkyl diquaternizing unit. Our aim is to combine very large, two-dimensional (2D) quadratic nonlinear optical (NLO) responses with reversible redox chemistry. The chromophores have been characterized as their PF_6^− salts by using various techniques including electronic absorption spectroscopy and cyclic voltammetry. Their visible absorption spectra are dominated by intense π → π^* intramolecular charge-transfer (ICT) bands, and all show two reversible diquat-based reductions. First hyperpolarizabilities β have been measured by using hyper-Rayleigh scattering with an 800 nm laser, and Stark spectroscopy of the ICT bands affords estimated static first hyperpolarizabilities β_0. The directly and indirectly derived β values are large and increase with the extent of π-conjugation and electron donor strength. Extending the quaternizing alkyl linkage always increases the ICT energy and decreases the E_(1/2) values for diquat reduction, but a compensating increase in the ICT intensity prevents significant decreases in Stark-based β_0 responses. Nine single-crystal X-ray structures have also been obtained. Time-dependent density functional theory clarifies the molecular electronic/optical properties, and finite field calculations agree with polarized HRS data in that the NLO responses of the disubstituted species are dominated by ‘off-diagonal’ β_(zyy) components. The most significant findings of these studies are: (i) β_0 values as much as 6 times that of the chromophore in the technologically important material (E)-4′-(dimethylamino)-N-methyl-4-stilbazolium tosylate; (ii) reversible electrochemistry that offers potential for redox-switching of optical properties over multiple states; (iii) strongly 2D NLO responses that may be exploited for novel practical applications; (iv) a new polar material, suitable for bulk NLO behavior

Deletion Mutants of VPg Reveal New Cytopathology Determinants in a Picornavirus
BACKGROUND: Success of a viral infection requires that each infected cell delivers a sufficient number of infectious particles to allow new rounds of infection. In picornaviruses, viral replication is initiated by the viral polymerase and a viral-coded protein, termed VPg, that primes RNA synthesis. Foot-and-mouth disease virus (FMDV) is exceptional among picornaviruses in that its genome encodes 3 copies of VPg. Why FMDV encodes three VPgs is unknown. METHODOLOGY AND PRINCIPAL FINDINGS: we have constructed four mutant FMDVS that encode only one VPG: either VPg(1), VPg(3), or two chimeric versions containing part of VPg(1) and VPg(3). All mutants, except that encoding only VPg(1), were replication-competent. Unexpectedly, despite being replication-competent, the mutants did not form plaques on BHK-21 cell monolayers. The one-VPg mutant FMDVs released lower amounts of encapsidated viral RNA to the extracellular environment than wild type FMDV, suggesting that deficient plaque formation was associated with insufficient release of infectious progeny. Mutant FMDVs subjected to serial passages in BHK-21 cells regained plaque-forming capacity without modification of the number of copies of VPg. Substitutions in non-structural proteins 2C, 3A and VPg were associated with restoration of plaque formation. Specifically, replacement R55W in 2C was repeatedly found in several mutant viruses that had regained competence in plaque development. The effect of R55W in 2C was to mediate an increase in the extracellular viral RNA release without a detectable increase of total viral RNA that correlated with an enhanced capacity to alter and detach BHK-21 cells from the monolayer, the first stage of cell killing. CONCLUSIONS: The results link the VPg copies in the FMDV genome with the cytopathology capacity of the virus, and have unveiled yet another function of 2C: modulation of picornavirus cell-to-cell transmission. A Reservoir Species for the Emerging Amphibian Pathogen Batrachochytrium dendrobatidis Thrives in a Landscape Decimated by Disease
Chytridiomycosis, a disease caused by the fungal pathogen Batrachochytrium dendrobatidis (Bd), is driving amphibian declines and extinctions in protected areas globally. The introduction of invasive reservoir species has been implicated in the spread of Bd but does not explain the appearance of the pathogen in remote protected areas. In the high elevation (>1500 m) Sierra Nevada of California, the native Pacific chorus frog, Pseudacris regilla, appears unaffected by chytridiomycosis while sympatric species experience catastrophic declines. We investigated whether P. regilla is a reservoir of Bd by comparing habitat occupancy before and after a major Bd outbreak and measuring infection in P. regilla in the field, monitoring susceptibility of P. regilla to Bd in the laboratory, examining tissues with histology to determine patterns of infection, and using an innovative soak technique to determine individual output of Bd zoospores in water. Pseudacris regilla persists at 100% of sites where a sympatric species has been extirpated from 72% in synchrony with a wave of Bd. In the laboratory, P. regilla carried loads of Bd as much as an order of magnitude higher than loads found lethal to sympatric species. Histology shows heavy Bd infection in patchy areas next to normal skin, a possible mechanism for tolerance. The soak technique was 77.8% effective at detecting Bd in water and showed an average output of 68 zoospores per minute per individual. The results of this study suggest P. regilla should act as a Bd reservoir and provide evidence of a tolerance mechanism in a reservoir species
Identifying Consensus Disease Pathways in Parkinson's Disease Using an Integrative Systems Biology Approach
Parkinson's disease (PD) has had six genome-wide association studies (GWAS) conducted as well as several gene expression studies. However, only variants in MAPT and SNCA have been consistently replicated. To improve the utility of these approaches, we applied pathway analyses integrating both GWAS and gene expression. The top 5000 SNPs (p<0.01) from a joint analysis of three existing PD GWAS were identified and each assigned to a gene. For gene expression, rather than the traditional comparison of one anatomical region between sets of patients and controls, we identified differentially expressed genes between adjacent Braak regions in each individual and adjusted using average control expression profiles. Over-represented pathways were calculated using a hyper-geometric statistical comparison. An integrated, systems meta-analysis of the over-represented pathways combined the expression and GWAS results using a Fisher's combined probability test. Four of the top seven pathways from each approach were identical. The top three pathways in the meta-analysis, with their corrected p-values, were axonal guidance (p = 2.8E-07), focal adhesion (p = 7.7E-06) and calcium signaling (p = 2.9E-05). These results support that a systems biology (pathway) approach will provide additional insight into the genetic etiology of PD and that these pathways have both biological and statistical support to be important in PD
